Study of Yeast Identification Using MALDI-TOF MS

چکیده

食品由来の酵母19菌種75株についてポテトデキストロース寒天培地, 25 °C,5日間培養した後, VITEK MSTM plusを用いてマススペクトルを取得し, SuperSpectrum (SSp) を作成した. 作成したSSpについて検証を行った結果, 同定の信頼性レベルはSchizosaccharomyces 属, Sterigmatomyces elviae, Starmerella apicola, S. etchellsii, Wickerhamomyces subpelliculosus, Zygosaccharomyces rouxii,Zygosaccharomyces bisporus では80 %以上であった. Z. bailii,Z. parabailii,Z. pseudobailii,Z. mellis, およびZ. siamensisは, それぞれのSSpでは判別することは困難であった. しかし,Z. pseudobailiiをZ. bailii complex, mellisおよびZ. siamensis をZ. mellis-siamensis complexとグループ化することで, それぞれのcomplexとしては同定することができた. 同定の信頼性レベルは検証に用いた酵母を培養した培地の種類により影響を受けた. また, Brettanomyces (Dekkera) 属では十分なマススペクトルが得られない菌種や, 主要ピークが類似した菌種で同定困難な事例が認められた. 今後, 抽出方法やSSpの作成方法を含め検討していく必要がある.
